Exploring the functions of miR164c/172a/319a/396a in the ectomycorrhizal formation of Populous tomentosa
要旨本文 MicroRNAs (miRNAs) play significant roles in regulatory mechanisms in mycorrhizal symbiosis. In the previous study, we identified the differentially expressed miRNAs (DE-miRNA) in roots of Populus tomentosa inoculated by Cenococcum geophilum by miR-seq. In this research, we selected four DE-miRNAs (miR164c, miR172a, miR319a and miR396a) as candidate miRNAs to reveal how miRNAs regulate ectomycorrhizal formation. We constructed the overexpressed, knocked-down and knocked-out transgenic mutants of P. tomemtosa for these four DE-miRNAs. At present, the overexpressed and knocked-down transgenic mutant lines were successfully regenerated. The knocked-out mutants of P. tomentosa by CRISPR/Cas9 system are being prepared. As the next step, we will investigate the regulating mechanisms of four DE-miRNAs by using the constructed transgenic mutants.
